I did some research and here are some info. regarding how to get to Big Sight for the Expo ~ I would suggest no matter where you are staying, try get to the Shimbashi Station 新橋駅 and then transfer to another private railway line called the Yurikamome Line. I think it is 800 Yen for a day pass. 10 stations and 22 minutes later, you will arrive at the front of the Big Sight. You can also take the Rinkai Line but it is underground. For sightseeing, Yurikamome is the best choice. You will get to see the Rainbow Bridge, Tokyo Bay and the Pacific Ocean.
The Big Sight is located on a piece of artifical land called Odaiba which was finally developed in the late 1990s. In this area, there are shopping, entertainment centres, museums and an amusement park. The only other building that I am interested to see there is the Fuji TV Station (cool design).
Here is the Tokyo Subway map. Shimbashi Station is located at the lower right side.

Please correct me if I am not right. Good luck!
